This stage marks the beginning of an accelerated aging phase, with profound impacts on the brain and the entire body. Although distinct in approach, both studies converge in their conclusions and highlight clear paths for prevention and early intervention.

Study 1: The Brain Begins to Age Faster at 44

A group of scientists led by researcher Lilianne R. Mujica-Parodi identified that the human brain enters a phase of accelerated aging starting at age 44, with progressive deterioration of neural networks until about age 67. After this period, the process tends to stabilize until around age 90.

How the Discovery Was Made

The findings were made possible through functional magnetic resonance imaging (fMRI) scans conducted on more than 19,000 individuals. The study showed that neuronal insulin resistance plays a central role in cognitive decline during this stage of life.

The researcher explains:

“We identified a critical midlife window, in which the brain begins to experience declining access to energy, but before irreversible damage occurs — essentially the ‘bend’ before the ‘break’.”

The Role of Brain Energy and Ketones

The study also evaluated the use of ketones, molecules that serve as an alternative energy source for the brain. Notably, the greatest benefits were observed in people aged 40 to 49, while older adults (60 to 79) did not show significant improvements.

Why Prevention Matters in Midlife

Co-author Botond Antal emphasized the importance of early preventive action:

“Instead of waiting for cognitive symptoms, which may not appear until substantial damage has already occurred, we can potentially identify people at risk through neurometabolic markers and intervene during this critical window.”


Study 2: The Human Body Ages in Two Peaks

A separate study, led by Professor Michael Snyder from the Center for Genomics and Personalized Medicine at Stanford University, found that human aging accelerates at two specific points in life: around age 44 and again around age 60.

Long-Term Monitoring of the Human Body

The research followed 108 volunteers aged 25 to 75, who provided blood, stool, saliva, skin, and mucosal samples over several years. Scientists analyzed more than 135,000 molecules and microorganisms, including RNA, proteins, metabolites, as well as bacteria, viruses, and fungi living in the gut and on the skin.

Professor Snyder summarized the findings:

“It turns out that your mid-40s is a time of dramatic change, just like the early 60s — and that’s true no matter which class of molecules you look at.”

Key Molecular Changes Identified

The molecular shifts observed include:

  • metabolism of alcohol, caffeine, and lipids;
  • early signs of cardiovascular disease;
  • changes in immune and kidney function;
  • aging of muscles and skin.

Men and Women Are Affected Alike

The first aging peak, detected in the 40s, was initially linked to perimenopause in women. However, the data showed that men experience similar changes during the same period, pointing to broader biological causes beyond female hormonal transitions.
